The *E. coli* tube detection system consists of a peptone-yeast extract base with lactose serving as the carbon source. The selective agents include bile salts, sodium lauryl sulfate, and other inhibitors of Gram-positive bacteria. The pH of the medium changes as a result of acidification caused by the metabolism of lactose. Bromcresol purple (BCP) is used as the pH indicator. The selectivity of the medium is enhanced through the use of a more concentrated inhibitory system and an incubation temperature of 44.5°C. As *E. coli* metabolizes the medium and produces acid, the color of the BCP changes from purple to yellow (the visual appearance of the tubes is similar to that of the Coliform tubes).
